摘要黑色素瘤是黑色素细胞转化而得的恶性肿瘤,目前寻找癌症的生物标记物非常困难。本课题体外培养正常的小鼠表皮黑色素细胞与黑色素瘤B16F10细胞,采用核磁共振氢谱技术,结合多维统计分析方法检测肿瘤细胞与正常细胞内源性小分子代谢物的区别,从而得出他们的代谢通路的区别。分析结果发现丙三醇、脂质、胆碱、β-羟丁酸、丙酮、肌酸、甘氨酸在癌细胞中明显增多,而亮氨酸、赖氨酸、精氨酸、乙酸盐、谷氨酸盐、葡萄糖、牛磺酸和氧化三甲胺相比于正常细胞有明显减少。这些显著性的变化与多种代谢途径相关,并可能成为黑色素瘤的潜在生物标记物。47859

Title   Metabolomics analysis of melanoma and normal melanocytes              

Abstract

Melanoma is a malignant tumor which transformed from melanocytes. It is difficult and challenging to find biomarkers of cancer from amount of compounds. In the study, we cultured normal melanocytes from mice and melanoma B16F10 in vitro at the same time and in the same conditions, and then used NMR-based metabolomics combined with multivariate date analysis to detect and identify endogenous molecular metabolites between the control and the melanoma groups. We also compared the difference of the pathways between them. We found that glycerol, lipids, choline, β-hydroxyburytate, acetone, creatine and glycine are increased statistically and significantly in the melanoma groups, while leucine, Ileleucine, lysine, arginine, acetate, glutamate, glucose, taurine and TMAO are statistically and significantly increased compared with the normal groups. These changes are associated with many biological pathways. They may become potential biomarkers for melanoma.
